PIWIL4 Antibody (YA4351) is a mouse-derived and non-conjugated IgG1 antibody, targeting to PIWIL4. It can be applicated for WB, IHC-P, FC, ELISA assays, in the background of human.

PIWIL4 plays a central role in spermatogenesis by repressing transposable elements and preventing their mobilization, which is essential for germline integrity (By similarity). It acts via the piRNA metabolic process, mediating transposable element repression during meiosis through complexes of piRNAs and Piwi proteins (By similarity). The PIWIL4-piRNA pathway silences active transposons in the nucleus by engaging nascent transposable element transcripts and directing DNA methylation (By similarity). Unlike PIWIL1 and PIWIL2, PIWIL4 lacks endonuclease activity (By similarity). It directly binds piRNAs (24–30 nt RNAs derived from transposons via a Dicer-independent mechanism) and associates with secondary antisense piRNAs, requiring PIWIL2/MILI for this interaction (By similarity). The piRNA process operates upstream of DNA methylation mediators (By similarity). PIWIL4 is key to the ping-pong amplification cycle, functioning as a 'slicer-incompetent' component that loads cleaved piRNAs from PIWIL2 ('slicer-competent') to target nuclear transposon loci (By similarity). It may induce histone H3 'Lys-9' methylation at specific loci (PubMed:17544373). Beyond germline functions, PIWIL4 regulates somatic activities (By similarity), including pancreatic beta cell function and insulin secretion (By similarity), and maintains retinal epithelial integrity via Akt/GSK3α/β signaling (PubMed:28025795). Overexpression promotes tumorigenesis by inhibiting apoptosis and enhancing proliferation (PubMed:22483988).
