Phospho-PKC zeta (Thr560) Antibody (YA162) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Phospho-PKC zeta (Thr560).

Background

PKC zeta is a calcium- and diacylglycerol-independent serine/threonine-protein kinase that operates in the PI3K pathway and MAP kinase cascade, playing roles in NF-kappa-B activation, mitogenic signaling, cell proliferation, polarity, inflammatory responses, and long-term potentiation (LTP) maintenance. In LPS-treated macrophages or mitogen-stimulated cells, it acts downstream of PI3K to activate the MAP2K1/MEK1-MAPK1/ERK2 cascade independently of RAF1. While involved in insulin-dependent AKT3 activation, it likely functions as an adaptor rather than a direct activator. Under insulin stimulation, PKC zeta may serve as a PI3K effector to promote SLC2A4/GLUT4 translocation and glucose uptake in adipocytes. In EGF-induced cells, it binds and activates MAP2K5/MEK5-MAPK7/ERK5 kinase-independently and stimulates the JUN promoter via MEF2C. Through SQSTM1/p62 interaction, it mediates IL-1 signaling and NF-kappa-B activation with RIPK1/TRAF6 adapters. For TNF-induced NF-kappa-B transactivation, it phosphorylates IKBKB to degrade NF-kappa-B inhibitors. In migrating astrocytes, PKC zeta forms a complex with PARD6A and is recruited by CDC42 to establish cell polarity alongside dynein. Partnered with FEZ1, it drives neuronal differentiation in PC12 cells. During inflammation, it is essential for Th2 differentiation (including IL production, JAK1 activation, and STAT6 nuclear translocation) and may contribute to Th2-dependent allergic airway inflammation (asthma). Within NF-kappa-B-mediated responses, it phosphorylates RELA at Ser-311 to counteract SETD6-mediated gene repression. In vitro, it phosphorylates VAMP2 and activates LRRK1 (which then phosphorylates RAB trafficking proteins). Additionally, PKC zeta participates in late-phase synaptic LTP in hippocampal CA1 cells and long-term memory maintenance.

  • Western blot analysis of extracts from Hela(lane 2(20μg) ) and Hela(lane 3(20μg), Calyculin A treated (HY-18983, 50nM, 3hrs) as indicated, using Phospho-PKC zeta (T560) (HY-P80280) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody (HY-P80280, 1/1000) and Loading control antibody (GAPDH, HY-P80954, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
