Phospho-Met(c-Met) Antibody (YA175) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Phospho-Met(c-Met).

Background

MET is a receptor tyrosine kinase that transduces signals from the extracellular matrix into the cytoplasm by binding to hepatocyte growth factor (HGF) ligand. It regulates various physiological processes, including proliferation, scattering, morphogenesis, and survival. Ligand binding induces autophosphorylation of MET on its intracellular domain, creating docking sites for downstream signaling molecules. Upon activation, MET interacts with the PI3-kinase subunit PIK3R1, PLCG1, SRC, GRB2, STAT3, or the adapter GAB1, leading to the activation of signaling cascades such as RAS-ERK, PI3K-AKT, or PLCgamma-PKC. The RAS-ERK pathway is associated with morphogenetic effects, while PI3K/AKT mediates prosurvival effects. During embryonic development, MET signaling contributes to gastrulation, neuronal precursor migration, angiogenesis, and kidney formation. In skeletal muscle development, it is essential for muscle progenitor cell migration and secondary myoblast proliferation (by similar). In adults, it participates in wound healing, organ regeneration, tissue remodeling, and promotes hematopoietic cell differentiation and proliferation. It may also regulate cortical bone osteogenesis (by similar). Additionally, MET acts as a receptor for Listeria monocytogenes internalin InlB, facilitating pathogen entry into cells.

  • Western blot analysis of extracts from A431 (lane 2(20μg) and A431(lane 3(40μg) using Phospho-Met (HY-P80512) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
