PD-L1 Antibody (YA235) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to PD-L1.

CD274 plays a critical role in the induction and maintenance of immune tolerance to self. As a ligand for the inhibitory receptor PDCD1/PD-1, it modulates the activation threshold of T-cells and limits T-cell effector responses. Through an unknown activating receptor, CD274 may costimulate T-cell subsets that predominantly produce interleukin-10 (IL10). It can also act as a transcription coactivator: in response to hypoxia, it translocates into the nucleus via interaction with phosphorylated STAT3 and promotes transcription of GSDMC, leading to pyroptosis. The PDCD1-mediated inhibitory pathway is exploited by tumors to attenuate anti-tumor immunity and escape immune system destruction, thereby facilitating tumor survival. The interaction with PDCD1/PD-1 inhibits cytotoxic T lymphocytes (CTLs) effector function (by similar). Blocking the PDCD1-mediated pathway reverses the exhausted T-cell phenotype and normalizes the anti-tumor response, providing a rationale for cancer immunotherapy (by similar).

  Western blot analysis of extracts from MDA-MB-231 (lane 2(20μg), THP-1(lane 3(20μg) and HEK293(lane 4(20μg) using PD-L1(HY-P80273) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
