P2RX7 Antibody (YA4449) is a mouse-derived and non-conjugated IgG2b antibody, targeting to P2RX7. It can be applicated for WB, IHC-P, FC, ELISA assays, in the background of human, mouse, rabbit.

P2RX7 is an ATP-gated nonselective transmembrane cation channel that requires high millimolar concentrations of ATP for activation. Upon ATP binding, it rapidly opens to allow the influx of small cations Na(+) and Ca²⁺, and the efflux of K(+). It also has the ability to form a large pore in the cell membrane, permitting the passage of large cationic molecules. In microglia, it may mediate NADPH transport across the plasma membrane. In immune cells, P2RX7 acts as a molecular sensor in pathological inflammatory states by detecting and responding to high extracellular ATP concentrations. In microglial cells, P2RX7 activation leads to the release of pro-inflammatory cytokines (e.g., IL-1beta and IL-18) via NLRP3 inflammasome and caspase-1 activation (cooperates with KCNK6 to activate NLRP3 inflammasome by similarity). It activates death pathways leading to apoptosis and autophagy, as well as pyroptosis (by similarity). Some variants exhibit ion channel activity but lack macropore function, rendering them non-functional channels.
