OTUD4 Antibody (YA4157) is a Mouse-derived and non-conjugated IgG1 monoclonal antibody, targeting to OTUD4.

OTUD4 is a deubiquitinase that hydrolyzes the isopeptide bond between the ubiquitin C-terminus and the lysine epsilon-amino group of target proteins. It may negatively regulate inflammatory and pathogen recognition signaling in innate immune responses. Upon phosphorylation at Ser-202 and Ser-204 residues via IL-1 receptor and Toll-like receptor signaling pathways, OTUD4 specifically deubiquitinates 'Lys-63'-polyubiquitinated MYD88 adapter protein, leading to downregulation of NF-kappa-B-dependent transcription of inflammatory mediators. Independently of its catalytic activity, OTUD4 acts as a scaffold for alternative deubiquitinases (e.g., USP7 and USP9X) to assemble specific deubiquitinase-substrate complexes, thereby stabilizing the alkylation repair enzyme ALKBH3 and promoting the repair of alkylated DNA lesions.
