NEDD9 is a scaffolding protein that plays a central coordinating role in tyrosine-kinase-based signaling related to cell adhesion. As a focal adhesion protein, it contributes to embryonic fibroblast migration (By similar). It may play a significant role in integrin beta-1 or B cell antigen receptor (BCR)-mediated signaling in B- and T-cells. Stimulation of integrin beta-1 leads to the recruitment of proteins such as CRKL and SHPTP2 to its tyrosine-phosphorylated form. It promotes lymphocyte adhesion and migration, ensuring proper migration to the spleen and other secondary lymphoid organs. Additionally, it regulates the organization of the T-cell F-actin cortical cytoskeleton and the centralization of T-cell receptor microclusters at the immunological synapse (By similar). NEDD9 negatively regulates cilia outgrowth in polarized cysts (By similar) and modulates cilia disassembly via AURKA-mediated phosphorylation of HDAC6 and subsequent deacetylation of α-tubulin. It positively regulates RANKL-induced osteoclastogenesis (By similar) and is essential for maintaining hippocampal dendritic spines in the dentate gyrus and CA1 regions, thereby contributing to spatial learning and memory (By similar).
