MuRF1 is an E3 ubiquitin ligase that mediates the ubiquitination and subsequent proteasomal degradation of CKM, GMEB1, and HIBADH. Under amino acid starvation, it regulates the proteasomal degradation of muscle proteins to provide amino acids for other organs while inhibiting de novo skeletal muscle protein synthesis. It also controls the degradation of cardiac troponin I/TNNI3 and likely other sarcomeric-associated proteins. Additionally, MuRF1 may influence striated muscle atrophy and hypertrophy by modulating an anti-hypertrophic PKC-mediated signaling pathway and regulates myofibril organization via TTN in muscle cells. by similar: may be involved in the degradation regulation of other muscle-related proteins.
