Description

MR1 Antibody (YA4415) is a mouse-derived and non-conjugated IgG2a antibody, targeting to MR1. It can be applicated for IHC-P, ICC/IF, FC, ELISA assays, in the background of human.

Background

MR1 is an antigen-presenting molecule specialized in displaying microbial pyrimidine-based metabolites to alpha-beta T cell receptors (TCR) on mucosal-associated invariant T (MAIT) cells. In complex with B2M, it preferentially presents riboflavin-derived metabolites to semi-invariant TRAV1.2 TCRs on MAIT cells, enabling immune surveillance of the microbial metabolome at mucosal epithelial barriers. Signature potent antigenic metabolites include 5-OE-RU and 5-OP-RU, formed by condensation of 5-A-RU with glyoxal or methylglyoxal byproducts, respectively. MR1 may also present microbial antigens to diverse TRAV1-2-negative MAIT cell subsets, facilitating recognition of microbes that do not synthesize riboflavin. Upon antigen recognition, it elicits rapid innate-type MAIT cell activation to eliminate pathogens by directly killing infected cells. During T cell development, MR1 drives thymic selection and post-thymic terminal differentiation of MAIT cells (by similar). Additionally, MR1 acts as an immune sensor of the cancer cell metabolome, potentially presenting tumor-specific or -associated metabolites to a 'pan-cancer' TCR on non-MAIT CD8-positive T cell clone (MC.7.G5), triggering T cell-mediated killing of various cancer types. Allele MR1*01 presents microbial-derived 5-OP-RU to semi-invariant TRAV1.2-TRAJ33-TRBV6.1 (A-F7) TCR on MAIT cells and captures nucleobase carbonyl adducts (e.g., M3Ade) generated during oxidative stress for recognition by MR1-restricted T cell clones with polyclonal TCR repertoires. Allele MR1*04 presents tumor-enriched pyridoxal to pan-cancer 7.G5 TCR on T cells, enabling preferential cancer cell recognition and potentially functioning as an alloantigen.
