MDA5 is an innate immune receptor that functions as a cytoplasmic sensor of viral nucleic acids, playing a pivotal role in detecting viral infections and triggering antiviral responses, including the induction of type I interferons and pro-inflammatory cytokines. Its ligands include mRNAs lacking 2'-O-methylation at their 5' cap and long dsRNA (>1 kb in length). Upon binding, MDA5 associates with the mitochondrial antiviral signaling protein (MAVS/IPS1), activating IKK-related kinases TBK1 and IKBKE, which phosphorylate interferon regulatory factors IRF3 and IRF7, leading to the transcription of antiviral immune genes such as IFN-alpha and IFN-beta. It detects members of the Picornaviridae family, including encephalomyocarditis virus (EMCV), mengo encephalomyocarditis virus (ENMG), and rhinovirus, as well as SARS-CoV-2. Additionally, MDA5 recognizes other viruses like dengue virus (DENV), West Nile virus (WNV), and reovirus, and responds to dsDNA-genome viruses such as vaccinia virus. It amplifies innate immune signaling by sensing RNA metabolites produced during viral infection by RNase L. MDA5 may also enhance natural killer cell activity and contribute to growth inhibition and apoptosis in certain tumor cell lines.
