Background

Leptin Receptor is a receptor for the hormone LEP/leptin (Probable). Upon ligand binding, it mediates the central and peripheral effects of LEP by activating various signaling pathways such as JAK2/STAT3 and MAPK cascade/FOS. In the hypothalamus, LEP acts as an appetite-regulating factor, reducing food intake and increasing energy expenditure by inducing anorexinogenic factors and suppressing orexigenic neuropeptides, while also regulating bone mass and hypothalamo-pituitary-adrenal hormone secretion (By similarity). In peripheral tissues, LEP enhances basal metabolism, influences reproductive function, modulates pancreatic beta-cell function and insulin secretion, exhibits pro-angiogenic properties, and impacts innate and adaptive immunity. Control of energy homeostasis and melanocortin production (via STAT3 signaling) is distinct from the regulation of NPY, fertility, growth, and glucose homeostasis. The receptor also modulates the counter-regulatory response to hypoglycemia by inhibiting parabrachial nucleus neurons and differentially affects T lymphocyte responses by regulating the proliferation of naive and memory T cells (By similarity). Additionally, Leptin Receptor may facilitate LEP transport across the blood-brain barrier, bind LEP, and mediate its endocytosis without inducing STAT3 phosphorylation or activation, while antagonizing Isoform A and isoform B-mediated LEP binding and endocytosis.
