ISG15 Antibody (YA333) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to ISG15.

ISG15 is a ubiquitin-like protein that plays a pivotal role in innate immune responses to viral infection through ISGylation (conjugation to target proteins) or as a free protein. ISGylation involves an enzymatic cascade (E1, E2, E3) that attaches ISG15 to lysine residues of targets like IFIT1, MX1/MxA, and PPM1B. For instance, ISGylation of the viral sensor IFIH1/MDA5 triggers oligomerization and immunity against coronaviruses, flaviviruses, and picornaviruses. ISG15 also modifies EIF2AK2/PKR (activating it), RIGI (inhibiting antiviral signaling), and EIF4E2 (enhancing translation suppression), while exhibiting broad antiviral activity against influenza A, HIV-1, and Ebola virus. The secreted form induces NK cell proliferation, acts as a neutrophil chemoattractant, and via ITGAL/ITGB2 receptor signaling, activates SRC kinases (LYN, HCK, FGR) to stimulate IFNG/IL10 secretion. by similar: ISG15 disrupts HIV-1 budding by blocking Gag-TSG101 interaction and inhibits Ebola VP40 ubiquitination by NEDD4 to suppress viral release.

  Western blot analysis of extracts from Hela (lane 2(20μg) , Hela (lane 3(40μg),using ISG15 Antibody (HY-P80194). Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
