Background

IRS2 is a signaling adapter protein involved in signal transduction from the insulin receptor (INSR) and insulin-like growth factor I receptor (IGF1R). It plays a crucial role in development, growth, glucose homeostasis, and lipid metabolism. Upon phosphorylation by the insulin receptor, IRS2 acts as a signaling scaffold, propagating insulin action by binding to SH2 domain-containing proteins such as the p85 regulatory subunit of PI3K, NCK1, NCK2, GRB2, or SHP2. Recruitment of GRB2 activates the guanine nucleotide exchange factor SOS1, triggering the Ras/Raf/MEK/MAPK signaling cascade (by similar). Activation of the PI3K/AKT pathway mediates most of insulin's metabolic effects in cells, while the Ras/Raf/MEK/MAPK pathway regulates gene expression and, in cooperation with PI3K, controls cell growth and differentiation. IRS2 positively regulates the Wnt/β-catenin signaling pathway by suppressing DVL2 autophagy-mediated degradation, promoting cell proliferation. It also contributes to cell cycle progression by enhancing the spindle assembly checkpoint (SAC) during M-phase. In macrophages, IL4-induced tyrosine phosphorylation of IRS2 recruits and activates phosphoinositide 3-kinase (PI3K).
