IKK gamma Antibody (YA346) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to IKK gamma.

Background

IKK gamma is the regulatory subunit of the IKK core complex, which phosphorylates inhibitors of NF-kappa-B, leading to the dissociation and degradation of the inhibitor/NF-kappa-B complex. Its binding to scaffolding polyubiquitin is crucial for IKK activation by multiple signaling receptor pathways. IKK gamma can recognize and bind both 'Lys-63'-linked and linear polyubiquitin upon cell stimulation, with a higher affinity for linear polyubiquitin. It may also play a role in NF-kappa-B-mediated protection from cytokine toxicity and is essential for viral activation of IRF3. Additionally, it is involved in the TLR3- and IFIH1-mediated antiviral innate immune response, requiring 'Lys-27'-linked polyubiquitination. Similar functions include serving as a mediator for HTLV-1 Tax oncoprotein activation of NF-kappa-B.

  • Immunohistochemical analysis of paraffin-embedded human Colon cancer‌ tissue using IKK gamma antibody was performed. The section was pretreated using high-temperature and high-pressure mediated EDTA antigen retrieval buffer (pH 9.0), for 5 minutes. The tissues were incubated with primary antibody (HY-P80415, 1:100 dilution) at room temperature for 60 minutes. Detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. The tissues were counterstained with hematoxylin and mounted with neutral balsam mounting medium.
  • Immunohistochemical analysis of paraffin-embedded human ovarian carcinoma tissue using IKK gamma antibody was performed. Then incubated with primary antibody (HY-P80415, 1:100 dilution) at room temperature for 60 minutes and HRP conjugated secondary antibody for 10 minutes. Fluorescence was then developed with TSA520 . The tissues were counterstained with DAPI and mounted with Anti-fade mounting medium.
