HIF1 alpha Antibody (YA4316) is a mouse-derived and non-conjugated IgG1 antibody, targeting to HIF1A. It can be applicated for WB, IHC-P, ICC/IF, ELISA assays, in the background of human, mouse, monkey.

HIF1A functions as a master transcriptional regulator of the adaptive response to hypoxia. Under hypoxic conditions, it activates the transcription of over 40 genes, including erythropoietin, glucose transporters, glycolytic enzymes, vascular endothelial growth factor, HILPDA, and other genes that enhance oxygen delivery or metabolic adaptation to hypoxia. HIF1A plays a critical role in embryonic vascularization, tumor angiogenesis, and ischemic disease pathophysiology. It heterodimerizes with ARNT (By similarity), binding to the hypoxia response element (HRE) core sequence 5'-TACGTG-3' in target gene promoters. Activation requires recruitment of coactivators like CREBBP and EP300, while interaction with NCOA1 or NCOA2 enhances its activity. Additionally, HIF1A's interaction with redox regulatory protein APEX1 activates its C-terminal transactivation domain (CTAD) and potentiates coactivator function. In neurons, HIF1A mediates mitochondrial distribution and transport during hypoxia. During microbial infection, HIF1A is essential for SARS-CoV-2-induced glycolysis in monocytes, promoting ACE2 and cytokine (e.g., IL1B, TNF, IL6, interferons) expression, thereby enhancing viral replication and inflammatory responses.
