Background

FOXP1 is a transcriptional repressor (PubMed:18347093, PubMed:26647308) that can synergistically repress transcription with CTBP1, though CTBP1 is not essential (By similarity). It plays a crucial role in lung epithelium specification and differentiation, cooperatively regulating lung secretory epithelial cell fate and regeneration with FOXP4 by restricting the goblet cell lineage program, potentially through AGR2 regulation. As a key transcriptional regulator of B-cell development, FOXP1 also modulates cardiac muscle cell proliferation and the columnar organization of spinal motor neurons, promoting lateral motor neuron column (LMC) and preganglionic motor column (PGC) formation for proper axon projections. It collaborates with other Hox proteins for segment-specific motor column generation and regulates PITX3 promoter activity, possibly promoting midbrain identity in stem cell-derived dopamine neurons. FOXP1 negatively regulates T follicular helper (T(FH)) cell differentiation and maintains hair follicle stem cell quiescence, likely via FGF18 regulation (By similarity). By inhibiting caspase-dependent apoptosis, it cooperates with NF-κB signaling to enhance B-cell expansion while repressing pro-apoptotic genes. FOXP1 binds CSF1R promoter elements to regulate monocyte differentiation and macrophage functions, requiring NCOR2 as a corepressor for CSF1R repression in monocytes (PubMed:15286807, PubMed:18347093, PubMed:18799727). It contributes to angiogenesis through endothelial cell proliferation, tube formation, and migration, potentially by suppressing SEMA5B (PubMed:24023716). FOXP1 negatively modulates androgen receptor signaling (PubMed:18640093) and acts as an AURKA-regulated transcriptional activator of the FBXL7 promoter (PubMed:28218735). In embryonic stem cells (ESCs), FOXP1 stimulates pluripotency-associated transcription factors while suppressing differentiation genes, exhibiting distinct DNA-binding specificity for sequences like 5'-CGATACAA-3' (PubMed:21924763) and promoting ESC self-renewal (By similarity).
