Background

EDD is an E3 ubiquitin-protein ligase involved in various protein quality control pathways in the cytoplasm and nucleus. It primarily functions as a ubiquitin chain elongator, extending pre-ubiquitinated substrates. As part of the N-end rule pathway, EDD ubiquitinates proteins with specific N-terminal residues that are destabilizing according to the N-end rule, leading to their degradation (by similar). EDD recognizes type-1 N-degrons containing positively charged amino acids (Arg, Lys, and His) (by similar). Together with UBR4, EDD participates in a cytoplasmic protein quality control pathway that prevents protein aggregation by catalyzing the assembly of heterotypic 'Lys-11'-/'Lys-48'-linked branched ubiquitin chains on aggregated proteins, facilitating their recognition by p97/VCP and subsequent proteasomal degradation. EDD also collaborates with ITCH to catalyze 'Lys-48'-/'Lys-63'-branched ubiquitination of TXNIP, promoting its degradation. In the nucleus, EDD mediates ubiquitination and degradation of unpaired transcription factors (those not assembled into functional multiprotein complexes). Additionally, EDD regulates mRNA maturation and transcriptional regulation by polyubiquitinating and activating CDK9. It plays a role in DNA damage response by suppressing RNF168 to prevent excessive spreading of ubiquitinated chromatin at damaged chromosomes. EDD positively regulates the canonical Wnt signaling pathway by stabilizing CTNNB1 and degrading TLE3 via 'Lys-48'-linked ubiquitination. It is essential for extraembryonic development (by similar) and maintains skeletal tissue homeostasis by inhibiting hedgehog (HH) signaling (by similar).
