Description

E-Cadherin Antibody (YA778) is a Mouse-derived and non-conjugated IgG1 monoclonal antibody, targeting to E-Cadherin.

Background

Cadherins are calcium-dependent cell adhesion proteins that preferentially interact homophilically between connected cells, contributing to the sorting of heterogeneous cell types. CDH1 regulates epithelial cell-cell adhesion, mobility, and proliferation. By interacting with AMOTL2 (by similar), it organizes radial actin fiber structures and mediates cellular responses to contractile forces by anchoring these fibers to CDH1 junction complexes at the membrane. During early desmosome formation, CDH1 facilitates recruitment of DSG2 and DSP to desmosomal plaques. It acts as a potent invasion suppressor and serves as a ligand for integrin α-E/β-7. E-Cad/CTF2 promotes non-amyloidogenic processing of Aβ precursors and strongly inhibits APP C99/C83 production. Additionally (Microbial infection), CDH1 functions as a receptor for Listeria monocytogenes via binding of bacterial InlA to mediate internalization.

  • Western blot analysis of extracts from A431(lane 2(20μg) and A431(lane 3(40μg) using E-Cadherin (HY-P80112) Mouse mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
  • Immunohistochemical analysis of paraffin-embedded Mouse colon tissue using E-Cadherin Antibody (YA778). The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P80112, 1/500) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
