Background

Desmoglein 2 is a core component of desmosomal cell-cell junctions, essential for positive regulation of cellular adhesion. It mediates the interaction between plaque proteins and intermediate filaments to reinforce intercellular adhesion and is critical for embryonic stem cell proliferation and viability in blastocysts, thereby supporting post-implantation embryonic development. The protein maintains pluripotency by modulating epithelial-mesenchymal transition (EMT)/mesenchymal-epithelial transition (MET) through sequestering CTNNB1 at cell-cell contacts, limiting its nuclear translocation and subsequent activation of CTNNB1/TCF-dependent transcription. It also enhances the pluripotency and multi-lineage differentiation capacity of hematopoietic stem cells and facilitates endothelial cell sprouting/elongation by coordinating cortical actin fiber-CDH5 junctional assembly. In kidney tubular epithelial cells, Desmoglein 2 may mitigate inflammatory infiltration and injury-induced apoptosis by preserving cell-cell adhesion and epithelial barrier integrity. Some functions are inferred from homology (By similarity).
