Background

DAP12 is an adapter protein that non-covalently associates with activating receptors on the surface of various immune cells to mediate signaling and cell activation following ligand binding. TYROBP is tyrosine-phosphorylated in the ITAM domain, leading to the activation of additional tyrosine kinases and subsequent cell activation. It also exhibits inhibitory roles in certain cells. DAP12 associates with activating receptors of the CD300 family to mediate cell activation and with CD200R family receptors (by similar) for the same purpose. It is essential for integrin-mediated neutrophil activation (by similar) and CLEC5A/MDL1 receptor-mediated myeloid cell activation. DAP12 interacts with NK cell receptors like KIR2DS2 and the KLRD1/KLRC2 heterodimer to activate NK cells and enhances the trafficking and surface expression of KIR2DS1, KIR2DS2, and KIR2DS4. It associates with SIRPB1 to activate myeloid cells (e.g., monocytes and dendritic cells) and with TREM1 to activate neutrophils and monocytes. Interaction with TREM2 upregulates CCR7 and promotes dendritic cell maturation and survival. DAP12 also mediates macrophage fusion into multinucleated giant cells via TREM2 and stabilizes the TREM2 C-terminal fragment (TREM2-CTF) to suppress pro-inflammatory cytokine release. In microglia, it collaborates with TREM2 for phagocytosis of apoptotic neurons (by similar) and with ITGAM/CD11B to regulate superoxide ion production. Additionally, DAP12 promotes pro-inflammatory responses in microglia after nerve injury, accelerating neuronal degeneration (by similar). It positively regulates IRAK3/IRAK-M kinase expression and IL10 production in liver dendritic cells, inhibiting their T cell stimulatory capacity (by similar), while negatively regulating B cell proliferation. DAP12 is also required for CSF1-mediated osteoclast cytoskeletal organization (by similar) and multinucleation during osteoclast development (by similar).
