Cytochrome P450 17A1 Antibody (YA478) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Cytochrome P450 17A1.

Cytochrome P450 17A1 is a cytochrome P450 monooxygenase involved in corticoid and androgen biosynthesis. It catalyzes the 17-alpha hydroxylation of C21 steroids, a common step in both pathways. A second oxidative step, required for androgen synthesis, involves acyl-carbon cleavage. The 17-alpha hydroxy intermediates serve as precursors of cortisol in the adrenal glucocorticoid biosynthesis pathway. This enzyme hydroxylates steroid hormones, pregnenolone, and progesterone to form 17-alpha hydroxy metabolites, followed by C17-C20 bond cleavage to produce C19 steroids such as dehydroepiandrosterone (DHEA) and androstenedione. It also exhibits 16-alpha hydroxylase activity, catalyzing the 16-alpha hydroxylation of 17-alpha hydroxy pregnenolone, followed by C17-C20 bond cleavage to form 16-alpha-hydroxy DHEA, and hydroxylates androgens, which is relevant for estriol synthesis. Mechanistically, it uses molecular oxygen, inserting one oxygen atom into the substrate and reducing the second to water, with electrons supplied by NADPH via cytochrome P450 reductase (CPR).

  Western blot analysis of extracts from Hela(lane 2(15μg) or lane 3(30μg) ) and Jurkat(lane 4(15μg) or lane 5(30μg) ) using Cytochrome P450 17A1(HY-P80449) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody (HY-P80449, 1/1000) and Loading control antibody (GAPDH, HY-P80954, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Rabbit/Mouse IgG-HRP Secondary Antibody (HY-P8001/HY-P8004, 1/10,000) was used for 1 hour at room temperature.
