Cullin 4A/4B Antibody (YA2583) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Cullin 4A/4B.

Cullin 4A/4B serves as the core component of multiple cullin-RING-based E3 ubiquitin-protein ligase complexes, facilitating ubiquitination and proteasomal degradation of target proteins. The functional specificity of these complexes is determined by variable substrate recognition subunits, with CUL4B potentially acting as a scaffold to position substrates and ubiquitin-conjugating enzymes. These complexes polyubiquitinate CDT1 and histones (H2A, H3, H4) in response to DNA damage, are recruited to UV-damaged chromatin via DDB2, and likely play roles in DNA repair/replication. Certain DCX complexes (containing TRPC4AP or DCAF12) participate in the DesCEND pathway by recognizing C-terminal degrons for targeted degradation. The DCX(AMBRA1) complex regulates G1/S transition by ubiquitinating phosphorylated cyclin-D (CCND1/2/3) and modulates CRL5 complexes via ELOC degradation. Cullin 4A/4B is essential for cyclin E (CCNE1/2) ubiquitination during G1 progression and regulates mTORC1 signaling through 26S proteasome-dependent interaction with MLST8. It collaborates with CUL4A in ribosome biogenesis.

  Western blot analysis was performed on extracts from Hela (lane 1, 15 μg), 293 (lane 2, 15 μg), 3T3 (lane 3, 15 μg), and Jurkat (lane 4, 15 μg) using Cullin 4A/4B Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Tubulin, HY-P80487, 1:60000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
