COPS8 Antibody (YA1679) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to COPS8.

Background

COPS8 is a component of the COP9 signalosome complex (CSN), which participates in various cellular and developmental processes. The CSN complex acts as a crucial regulator of the ubiquitin (Ubl) conjugation pathway by mediating the deneddylation of cullin subunits in SCF-type E3 ligase complexes (e.g., SCF, CSA, or DDB2), thereby reducing their Ubl ligase activity. Additionally, through its association with CK2 and PKD kinases, the complex is involved in the phosphorylation of p53/TP53, c-jun/JUN, IkappaBalpha/NFKBIA, ITPK1, and IRF8/ICSBP. CSN-dependent phosphorylation of TP53 and JUN promotes and protects their degradation by the Ubl system, respectively.

  • Western blot analysis of extracts from Hela(lane 2(20ug) ,HepG2(lane 3(20ug) and NIH/3T3(lane 4(20ug)using COPS8 Antibody (HY-P81934) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
  • Immunohistochemical analysis of paraffin-embedded Rat brain tissue using COPS8 Antibody (YA1679). The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P81934, 1/100) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
