Clusterin Antibody (YA5546) is a mouse-derived and non-conjugated IgG1 monoclonal antibody, targeting to Clusterin. It can be applicated for WB, ICC/IF, ELISA assays, in the background of human, mouse, rat.

Clusterin functions as an extracellular chaperone that prevents aggregation of non-native proteins and inhibits stress-induced aggregation of plasma proteins. In vitro, it suppresses amyloid fibril formation by APP, APOC2, B2M, CALCA, CSN3, SNCA, and aggregation-prone LYZ variants without requiring ATP. It maintains partially unfolded proteins in a refolding-competent state for other chaperones like HSPA8/HSC70 but lacks independent refolding activity. Binding to cell surface receptors triggers internalization and lysosomal/proteasomal degradation of client complexes. Clusterin also protects against apoptosis and complement-mediated cytolysis by inhibiting MAC assembly via blocking C9 polymerization. Intracellular forms interact with ubiquitin and SCF E3 ligases to promote degradation of targets (e.g., COMMD1, IKBKB) and modulate NF-kappa-B activity. Mitochondrial Clusterin suppresses BAX-dependent cytochrome c release and stabilizes membrane integrity via HSPA5 binding. The secreted form does not affect caspase/BAX-mediated apoptosis or TNF-induced NF-kappa-B but regulates neuronal differentiation through STMN3 interaction (by similar) and may clear immune complexes during cell injury (by similar). Paradoxically, it can also promote cell death by activating BAX via BCL2L1 interaction.
