Background

Clock is a transcriptional activator that serves as a core component of the circadian clock, an internal time-keeping system that generates approximately 24-hour rhythms in gene expression, metabolism, and behavior. Derived from the Latin roots 'circa' (about) and 'diem' (day), it regulates diverse physiological functions including metabolism, sleep, blood pressure, and immune responses. The clock consists of a central pacemaker in the suprachiasmatic nucleus (SCN) of the brain and peripheral clocks in tissues, both synchronized by environmental Zeitgebers ('time-givers'), primarily light for the SCN. The molecular mechanism involves a transcription-translation feedback loop (TTFL) where CLOCK-BMAL1 heterodimers (positive limb) activate E-box-containing genes, while PER/CRY proteins (negative limb) inhibit this activity. CLOCK also exhibits acetyltransferase activity, rhythmically modifying histones and nonhistone proteins (e.g., BMAL1) to regulate chromatin accessibility. Disrupted circadian rhythms contribute to diseases like cancer and metabolic disorders. By similarity: Drives blood pressure rhythms via transcriptional activation of ATP1B1.
