CASP-7 is a thiol protease involved in various programmed cell death processes, including apoptosis, pyroptosis, and granzyme-mediated cell death, by cleaving target proteins. It exhibits a strong preference for the Asp-Glu-Val-Asp (DEVD) consensus motif but also accommodates non-canonical sequences. As an effector caspase, CASP-7 participates in the execution phase of apoptosis upon activation by initiator caspases (e.g., CASP8, CASP9, or CASP10), cleaving substrates such as CLSPN, PARP1, PTGES3, and YY1. Compared to CASP3, it has a more restricted substrate profile. During bacterial infection, CASP-7 regulates inflammatory responses by cleaving and activating SMPD1 to promote membrane repair. In intestinal epithelial cells, CASP-1-activated CASP-7 is secreted extracellularly, where it stimulates SMPD1-derived ceramide production to counteract GSDMD pore activity (by similar). In hepatocytes, GZMB-activated CASP-7 is secreted to neutralize PRF1 pores via SMPD1 (by similar). Additionally, CASP-7 suppresses type I interferon production during viral infection by cleaving antiviral proteins CGAS, IRF3, and MAVS (by similar), activates SREBPs and phospholipid scramblases (XKR4/XKR8/XKR9) (by similar), and inhibits Kaposi sarcoma-associated herpesvirus lytic gene expression by cleaving ORF57.
