CIB1 is a calcium-binding protein involved in regulating various cellular processes, including cell differentiation, division, proliferation, migration, thrombosis, angiogenesis, cardiac hypertrophy, and apoptosis. It negatively regulates the thrombopoietin-mediated signaling pathway during bone marrow megakaryocyte differentiation and participates in the endomitotic cell cycle. CIB1 inhibits α-IIb/β3 integrin activation in thrombin-stimulated megakaryocytes, preventing platelet aggregation. It upregulates PTK2/FAK1 activity and facilitates its recruitment to focal adhesions, playing a key role in their formation. CIB1 promotes CDC42-dependent cell migration on fibronectin, negatively regulated by PAK1, and suppresses stress-activated MAPK signaling. It modulates microtubule dynamics during neuronal development by inhibiting STMN2-mediated depolymerization. Additionally, CIB1 activates the calcineurin/NFAT pathway to induce cardiomyocyte hypertrophy and stimulates endothelial cell proliferation and migration in ischemia-induced angiogenesis via PAK1 and ERK1/ERK2 signaling. It forms a complex with TMC6/EVER1 and TMC8/EVER2 in lymphocytes and keratinocytes, stabilizing their levels and enhancing intrinsic immunity against human β-papillomaviruses (HPVs). by similar: CIB1 also mediates PKD/PRKD2-induced VEGFA secretion, regulating angiogenesis and tumor growth.
