Description

Background

CD209 is a pathogen-recognition receptor expressed on the surface of immature dendritic cells (DCs) and involved in the initiation of primary immune responses. It mediates the endocytosis of pathogens by binding to mannose-like carbohydrates, followed by degradation in lysosomal compartments. The receptor returns to the cell membrane surface, and pathogen-derived antigens are presented to resting T-cells via MHC class II proteins to initiate adaptive immune responses. On DCs, CD209 acts as a high-affinity receptor for ICAM2 and ICAM3, potentially mediating the transendothelial migration of DC precursors from blood to tissues by binding endothelial ICAM2. It also regulates DC-induced T-cell proliferation by binding to ICAM3 on T-cells within the immunological synapse. CD209 serves as an attachment receptor for various viruses, including HIV-1, HIV-2, Ebolavirus, Cytomegalovirus, HCV, Dengue virus, Measles virus, Herpes simplex virus 1, Influenzavirus A, SARS-CoV, Japanese encephalitis virus, Lassa virus, Marburg virus, Respiratory syncytial virus, Rift Valley fever virus, Uukuniemi virus, and West Nile virus. Additionally, CD209 likely recognizes high mannose N-linked oligosaccharides in bacterial pathogen antigens, such as Leishmania pifanoi LPG, Helicobacter pylori LPS (Lewis-x antigen), Klebsiella pneumoniae LPS (mannose), Mycobacterium tuberculosis ManLAM (di-mannose and tri-mannose), and Schistosoma mansoni SEA (Lewis-x antigen), in a calcium-dependent manner. Dendritic cell recognition of M. tuberculosis occurs partially through CD209.
