Description

CD16 Antibody (YA5365) is a mouse-derived and non-conjugated monoclonal antibody, targeting to CD16. It can be applicated for FC assays, in the background of human.

Background

CD16 is a receptor for the invariable Fc fragment of immunoglobulin gamma (IgG), optimally activated upon binding clustered antigen-IgG complexes on cell surfaces, triggering antibody-dependent cellular cytotoxicity (ADCC) and lysis of antibody-coated cells. It does not bind free monomeric IgG, preventing inappropriate effector cell activation in the absence of antigen. On natural killer (NK) cells, it mediates IgG effector functions by binding antigen-IgG complexes generated during infection, inducing NK cell-dependent cytokine production and degranulation to limit viral load and spread. It is involved in generating memory-like adaptive NK cells capable of producing high levels of IFNG and efficiently eliminating virus-infected cells via ADCC. CD16 regulates NK cell survival and proliferation, particularly by preventing apoptosis of NK cell progenitors. As an Fc-binding subunit, it associates with CD247 and/or FCER1G adapters to form functional signaling complexes. Upon antigen-IgG complex engagement, it triggers phosphorylation of immunoreceptor tyrosine-based activation motif (ITAM)-containing adapters, activating phosphatidylinositol 3-kinase signaling and sustaining elevated intracellular calcium to drive NK cell activation. ITAM-dependent signaling coupled with receptor phosphorylation by PKC mediates robust calcium flux, leading to pro-inflammatory cytokine production, whereas in the absence of phosphorylation, it primarily activates phosphatidylinositol 3-kinase signaling, resulting in degranulation. It also costimulates NK cells and triggers target cell lysis independently of IgG binding. CD16 mediates the antitumor effects of therapeutic antibodies, triggering TNFA-dependent ADCC of IgG-coated tumor cells upon ligation on monocytes. It enhances ADCC in response to afucosylated IgGs. In dengue virus infection, it contributes to pathogenesis via antibody-dependent enhancement (ADE), where secondary infection elevates afucosylated non-neutralizing IgG1 levels that react with viral envelope/E protein. These antigen-IgG1 complexes bind FCGR3A with high affinity, facilitating viral entry into myeloid cells and subsequent replication.
