BMAL1 Antibody (YA581) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to BMAL1.

BMAL1 is a transcriptional activator that serves as a core component of the circadian clock, an internal timekeeping system generating approximately 24-hour rhythms in gene expression to regulate metabolism and behavior. Derived from the Latin 'circa' (about) and 'diem' (day), it governs diverse physiological processes including sleep, body temperature, blood pressure, and endocrine/immune functions. The clock consists of a central pacemaker in the suprachiasmatic nucleus (SCN) and peripheral oscillators in tissues, synchronized by Zeitgebers like light. BMAL1 heterodimerizes with CLOCK or NPAS2 to form the positive limb of the transcription-translation feedback loop (TTFL), activating E-box (5'-CACGTG-3')-containing clock genes (e.g., PER1/2/3, CRY1/2) and metabolic genes. These repressors then inhibit BMAL1 activity, completing the negative feedback. BMAL1 also regulates myogenesis (via Wnt signaling), pancreatic β-cell function (through NFE2L2-mediated antioxidant responses), and mTORC1 pathway suppression. It controls monocyte recruitment rhythms, ovarian steroidogenesis (e.g., CYP19A1), and hippocampal neurogenesis by modulating neural stem cell cycle entry. The CLOCK-BMAL1 heterodimer preferentially binds 5'-CACGTGA-3' and non-canonical E-box variants (e.g., 5'-AACGTGA-3'), driving rhythmic chromatin opening. In infection contexts, BMAL1 may post-transcriptionally regulate ACE2 and interferon-stimulated genes to influence SARS-CoV-2 entry. By similarity, it limits CD274 expression in macrophages (via PKM2) to combat sepsis and promotes skeletal muscle metabolic efficiency by activating DGAT2 and COQ10B.

  Western blot analysis of extracts from Hela(lane 2(20μg) , Hela(lane 3(40μg) and NIH3T3(lane 4(20ug) , NIH3T3(lane 5(40ug) using BMAL1(HY-P80033 Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  Immunocytochemistry analysis of Hela cells labeling BMAL1 with BMAL1 Antibody (HY-P80033) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with BMAL1 Antibody (HY-P80033) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 594-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8003, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).
