ATRX Antibody (YA828) is a Mouse-derived and non-conjugated IgG1 monoclonal antibody, targeting to ATRX.

Background

ATRX is a protein involved in transcriptional regulation and chromatin remodeling. It facilitates DNA replication in various cellular contexts and is essential for efficient replication at specific genomic loci. ATRX binds to DNA tandem repeats in telomeres and euchromatin, as well as to DNA quadruplex structures in vitro. It may stabilize G-rich regions by remodeling G4 DNA and incorporating H3.3-containing nucleosomes into regular chromatin structures. As the catalytic component of the ATRX:DAXX chromatin remodeling complex, it exhibits ATP-dependent DNA translocase activity, enabling replication-independent deposition of histone H3.3 in pericentric repeats and telomeres outside S-phase, along with in vitro remodeling of H3.3 nucleosomes. Its heterochromatin targeting likely involves combinatorial recognition of histone H3 modifications (e.g., H3K9 and H3K4 methylation states) and interaction with CBX5. In embryonic stem cells, ATRX maintains telomere integrity, potentially by recruiting CBX5. Its role in regulating telomeric repeat-containing RNA (TERRA) transcription remains debated, with some studies indicating transcriptional repression in stem cells. ATRX negatively regulates chromatin incorporation of repressive histone MACROH2A1, particularly at telomeres. It also contributes to allele-specific expression at the imprinted IGF2/H19 locus, where it facilitates maternal allele-specific occupancy of SMC1 and CTCF in the H19 imprinting control region (ICR) and establishes histone modifications. Additionally, ATRX binds zinc-finger genes with atypical chromatin signatures and modulates their H3K9me3 levels (By similarity: forms a complex with ZNF274, TRIM28, and SETDB1 to deposit/maintain H3K9me3 at zinc-finger gene 3' exons).
