ATG4A Antibody (YA4393) is a mouse-derived and non-conjugated IgG2b antibody, targeting to ATG4A. It can be applicated for ICC/IF, FC, ELISA assays, in the background of human.

ATG4A is a cysteine protease that plays a crucial role in autophagy by mediating both proteolytic activation and delipidation of ATG8 family proteins. Its protease activity cleaves the C-terminal amino acid of ATG8 proteins to expose a glycine residue, which is essential for their conjugation to phosphatidylethanolamine (PE) and membrane insertion. The preferred substrates are GABARAPL2, followed by MAP1LC3A and GABARAP. Additionally, ATG4A counteracts ATG8ylation by acting as a deubiquitinating-like enzyme, removing ATG8 conjugated to proteins like ATG3. It also exhibits stronger delipidation activity compared to ATG4B, catalyzing the removal of PE from ATG8 proteins during autophagy. Beyond its protease function, ATG4A contributes to mitophagy by regulating ATG9A trafficking to mitochondria and promoting phagophore-endoplasmic reticulum contacts, independent of its protease activity and ATG8 proteins.
