Background

ATF5 is a transcription factor that either stimulates or represses gene transcription by binding to different DNA regulatory elements, including the cAMP response element (CRE), ATF5-specific response element (ARE), and amino acid response element (AARE). It plays a critical role in cell survival, proliferation, and differentiation, often in a cell type-dependent manner. Its transcriptional activity is enhanced by CCND3 and slightly inhibited by CDK4. ATF5 is a key regulator of cerebral cortex formation, maintaining proliferation and blocking differentiation of cerebral cortical neuroprogenitor cells (By similar). It also participates in pathways where SHH promotes cerebellar granule neuron progenitor cell proliferation (By similar) and is essential for the survival of mature olfactory sensory neurons (OSNs), directing OSN-specific gene expression (By similar). ATF5 may be involved in osteogenic differentiation and promotes cell proliferation and survival by synergistically inducing EGR1 expression with ELK1. Once acetylated by EP300, it binds to ARE sequences on target gene promoters, such as BCL2 and EGR1. It exhibits an anti-apoptotic role in certain cell types through BCL2 regulation (By similar). ATF5 cooperates with NR1I3/CAR to activate CYP2B6 transcription in the liver but represses CRE-dependent transcription and inhibits proliferation by arresting cells at the G2/M phase in hepatic cells. It may act as a negative regulator of the IL1B transduction pathway in the liver and, upon IL1B stimulation, cooperates with NLK to enhance the transactivation activity of C/EBP subfamily members. Beyond its transcription factor role, ATF5 acts as a cofactor for CEBPB to activate CEBPA and promote adipocyte differentiation. Additionally, it regulates centrosome dynamics in a cell-cycle- and centriole-age-dependent manner, forming a 9-foci symmetrical ring scaffold around the mother centriole to control centrosome function and centriole-pericentriolar material interactions.
