ARFGAP1 is a GTPase-activating protein (GAP) for ADP ribosylation factor 1 (ARF1). It plays a role in membrane trafficking and/or vesicle transport by promoting the hydrolysis of ARF1-bound GTP, which is essential for the dissociation of coat proteins from Golgi-derived membranes and vesicles, a prerequisite for vesicle fusion with target compartments. ARFGAP1 likely regulates ARF1-mediated transport through its interaction with KDELR proteins and TMED2. Overexpression of ARFGAP1 induces the redistribution of the entire Golgi complex to the endoplasmic reticulum, similar to the effect observed when ARF1 is deactivated. Its activity is stimulated by phosphoinositides and inhibited by phosphatidylcholine (by similarity).
