Background

Aly (ALYREF/THOC4) functions as an mRNA export adaptor and core component of the transcription/export (TREX) complex, coupling mRNA transcription, processing with nuclear export (by similar). It specifically binds spliced mRNAs (but not unspliced pre-mRNAs) and mediates mRNA export to the cytoplasm via the TAP/NXF1 pathway (by similar). Aly plays a pivotal role in mRNP recognition and mRNA packaging by bridging the EJC (exon-junction complex)-bound mRNP with the TREX core complex (by similar). Additionally, it recognizes and binds m5C-modified mRNAs to facilitate their nucleocytoplasmic transport, and acts as a chaperone to promote dimerization of bZIP domain-containing transcription factors for transcriptional activation (by similar). (Microbial infection) The TREX complex is essential for Kaposi’s sarcoma-associated herpesvirus (KSHV) intronless mRNA export and infectious virion production, where Aly mediates TREX recruitment to viral mRNAs (by similar).
