Description

5-Fluoroorotic acid monohydrate is the monohydrate salt form of 5-Fluoroorotic acid (HY-W016819). 5-Fluoroorotic acid monohydrate is the inhibitor for thymidylate synthase that acts as a selective agent in yeast molecular genetics. 5-Fluoroorotic acid monohydrate exhibits antimalarial activity[1][2][3].

In Vitro

5-Fluoroorotic acid monohydrate (1 mg/mL, 4-7 days) inhibits thymidylate synthase, selectively inhibits the growth of Ura+ cells with the presence of UMP synthase, and can be used for screening of Ura- mutated yeast (such as Saccharomyces cerevisiae)[1][3].
5-Fluoroorotic acid monohydrate (1 nM-100 μM, 48 h) exhibits antimalarial activity against Plasmodium falciparum with an IC50 of 6.0 nM. 5-Fluoroorotic acid monohydrate exhibits relatively lower cytotoxicity in mammalian cell HT-1080 with an IC50 of 10 μM[4][5].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Cell Proliferation Assay[4]

Cell Line: HT-1080
Concentration: 1 nM-100 μM
Incubation Time: 48 h
Result: Inhibited cell proliferation.
In Vivo

5-Fluoroorotic acid monohydrate (0.2-5 mg/kg, ip, every 4 hours for 3 days) suppresses malarial parasitemia, exhibits toxicity with the absence of uridine (at a dose of 5 mg/kg) in P. yoelii-infected mouse models[5].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model: P. yoelii-infected mouse models[5]
Dosage: 0.2-5 mg/kg
Administration: ip, every 4 hours for 3 days)
Result: Eliminated malarial parasites.
Caused weight loss, diarrhea, decreased numbers of leukocytes, and increased mortality (5 mg/kg).

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
