Description

XQ2B is a specific cGAS inhibitor targeting protein-DNA interaction and phase separation. XQ2B markedly reduces ISD-induced Autophagy. XQ2B inhibits herpes simplex virus 1 (HSV-1)-induced antiviral immune responses and enhances HSV-1 infection[1].

In Vitro

XQ2B (10 μM; 24 h) inhibits the expression of Ifnb1, Cxcl10 and Il6 in Trex1-/- BMDMs[1].

In Vivo

XQ2B (10 mg/kg; i.v.; 3  h) attenuates the host innate antiviral (HSV-1) response in WT mice[1].
XQ2B (10 mg/kg; i.v.; every other day; 7 days) suppresses systemic inflammation in Trex1-/- mice[1].

Animal Model: C57BL/6 Trex1-/- male mice (8-week-old) with autoimmune disease model[1]
Dosage: 10 mg/kg
Administration: Intravenous injection, every other day, 7-day cycle for evaluating anti-inflammatory effect and 11 - day cycle for survival analysis
Result: Attenuated the inflammation in heart, stomach, tongue, kidney, and skeletal muscle of Trex1-/- mice compared to untreated Trex1-/- mice (the 7-day cycle experiment).
Suppressed the mRNA level of Ifnb1, Cxcl10 and Il6 caused by Trex1 deficiency.
Showed no effect on mouse survival during the test period (the 11-day cycle experiment).
