Description

SPiDER-βGal-2 is a fluorogenic β-galactosidase substrate and suitable for labeling live cells in culture and in living tissues. SPiDER-βGal-2 can generate quinone methide through enzymatic reaction and form covalent bonds with nucleophilic functional groups of proteins near the molecule, so that the fluorescent product is fixed in living cells (Ex/Em = 493, 525 nm/560 nm)[1][2].

In Vitro

1. Solution preparation
1.1 Preparation of stock solution
Solvent: DMSO
Concentration: 1 mM (optimized according to the experiment).
Storage: Store at -20°C or -80°C in dark after aliquoting. Avoid repeated freezing and thawing.
1.2 Preparation of working solution
Dilute to 1 µM with Hanks’ HEPES buffer (optimized according to the experiment).
Note: The working solution should be prepared and used immediately. Keep it away from light.

2. Cell staining
2.1 Prepare cells for the assay.
2.2 Discard the culture medium and wash the cells with Hanks’ HEPES buffer twice.
3. Add SPiDER-βGal-2 (1 µM) working solution.
4. Incubate at 37°C for 15 minutes (optimized according to the experiment).
5. Observe the cells under a fluorescence microscope or by a flow cytometer (Ex/Em = 500-540/560 nm).
