Background

RIB/ALK-4 is a transmembrane serine/threonine kinase activin type-1 receptor that forms an activin receptor complex with activin receptor type-2 (ACVR2A or ACVR2B). It transduces activin signals from the cell surface to the cytoplasm, regulating various physiological and pathological processes, including neuronal differentiation and survival, hair follicle development and cycling, pituitary FSH production, wound healing, extracellular matrix production, immunosuppression, and carcinogenesis. Activin is also implicated in paracrine or autocrine roles during ovarian follicular development. Within the receptor complex, type-2 receptors (ACVR2A and/or ACVR2B) serve as primary activin receptors, while type-1 receptors like ACVR1B act as downstream transducers. Activin binds to type-2 receptors at the plasma membrane, activating their serine-threonine kinase activity, which then phosphorylates and activates type-1 receptors such as ACVR1B. Activated type-1 receptors phosphorylate SMAD2 and SMAD3 at C-terminal serine residues, leading to their release into the cytoplasm, where they form a complex with SMAD4. This SMAD complex translocates to the nucleus to mediate activin-induced transcription. Inhibitory SMAD7, recruited to ACVR1B via FKBP1A, blocks activin signaling by preventing SMAD2/SMAD3 association with the receptor complex. Additionally, inhibin-B binding via the IGSF1 inhibin coreceptor antagonizes activin signaling. ACVR1B also phosphorylates TDP2.
