HRG/HPRG Protein Protein, Human (HEK293, His) is the recombinant human-derived HRG/HPRG protein, expressed by HEK293, with C-His tag.

Background

HRG/HPRG is a plasma glycoprotein that binds various ligands, including heme, heparin, heparan sulfate, thrombospondin, plasminogen, and divalent metal ions. It binds heparin and heparin/glycosaminoglycans in a zinc-dependent manner and interacts with heparan sulfate on the surface of endothelial cells in the liver, lung, kidney, and heart. Additionally, it binds N-sulfated polysaccharide chains on liver endothelial cells and inhibits rosette formation. Functioning as an adapter protein, HRG/HPRG regulates processes such as immune complex and pathogen clearance, cell chemotaxis, adhesion, angiogenesis, coagulation, and fibrinolysis. It enhances phagocytosis of necrotic cells via a heparan sulfate-dependent pathway, modulated by ligands like heparin and zinc ions. HRG/HPRG also binds IgG subclasses with kappa and lambda light chains, regulating their clearance and preventing insoluble immune complex formation. It tethers plasminogen to cell surfaces, binds T-cells to alter morphology, and modulates angiogenesis by counteracting the CD6-mediated antiangiogenic effects of THBS1 and THBS2. As a regulator of VEGF signaling, it inhibits endothelial cell motility by reducing VEGF-induced PXN/paxillin-ILK/integrin-linked protein kinase complex formation and suppressing tyrosine phosphorylation of focal adhesion kinases and alpha-actinins. Furthermore, HRG/HPRG plays roles in tumor angiogenesis and immune surveillance, normalizing tumor vasculature and promoting antitumor immunity by polarizing tumor-associated macrophages, thereby reducing tumor growth and metastasis.
