Description

RALA peptide is a cationic amphiphilic delivery agent targeting cell membranes, which forms nanoparticles through electrostatic interactions with anionic drugs. RALA peptide has pH-responsive binding properties, which can enhance the α-helical conformation in an acidic environment and destroy the endosomal membrane, promote the release of drugs into the cytoplasm, and exert efficient intracellular delivery activity. RALA peptide can be used in cancer research (enhancing the activity of bisphosphonates against prostate cancer and breast cancer cells) and bone tissue engineering (promoting osteoblast collagen deposition and extracellular matrix mineralization)[1][2][3].

In Vitro

Nanoparticles formed by RALA peptide (mass ratio of 10:1 to bisphosphonate; 6 h in vitro treatment and 72 h culture) and bisphosphonate significantly enhances the antiproliferative activity against PC3 prostate cancer cells and MDA-MB-231 breast cancer cells[1].
Nanoparticles formed by RALA peptide (mass ratio of 5:1 to α-TCP; 24 h in vitro transfection and 28 d culture) combined with α-TCP, promotes the expression and deposition of type I collagen in MG-63 osteoblasts and enhances the extracellular matrix mineralization of porcine mesenchymal stem cells[2][3].
After RALA peptide (mass ratio of 5:1 to α-TCP; cultured in vitro for 2-4 weeks) deliveres to mesenchymal stem cells, significantly upregulates the expression of BMP-2 and core osteogenic genes and accelerated the mineralization process in the engineered hydrogel[3].

In Vivo

RALA peptide (10 μg alendronate/time; intratumoral injection; 3 times/week; for 3 weeks) significantly delays tumor growth in PC3 tumor-bearing mouse model and prolongs the median survival time of mice to 26 days (16 days in control group), confirming that RALA/alendronate nanoparticles enhance the anti-tumor effect in vivo by enhancing drug delivery[1].
RALA peptide (composite α-TCP nanoparticles; subcutaneous implantation; single implantation; in vivo implantation for 8 weeks) promotes osteogenic differentiation and vascularization to achieve efficient bone tissue regeneration in Balb/C nude mouse model, induces bone tissue formation in hydrogel loaded with mesenchymal stem cells, without significantly different in mineralization volume from that in the growth factor delivery group, indicating that RALA-mediated α-TCP delivery can replace growth factors to promote bone regeneration in vivo[3].
