Description

OG 488, SE (Oregon green 488 succinimidyl ester), a fluorescent pH indicator, has many applications in biochemistry and neurosciences[1][2].

In Vitro

Particle Preparation[1]
1. Dilute 250 µL of a 5.75% polystyrene particle suspension with 0.1 M sodium bicarbonate buffer (pH 8.3) to a total volume of 1.5 mL, achieving a 6-fold dilution. Then, place the mixture in an ice bath and sonicate for 5 minutes.
2. While stirring gently, slowly add 1.5 mL of 300 µM OG 488, SE labeling reagent and an equal amount of 0.1 M sodium bicarbonate buffer (pH 8.3) to the solution.
3. After gently stirring, let the mixture incubate in a dark environment at room temperature for 2 hours.
4. Next, take 15 µL of 40 µM Texas Red and SE labeling reagent dissolved in dimethylformamide and carefully add it to the particle solution under ultrasonic treatment to ensure even dispersion.
5. After incubation, slowly centrifuge the particles at 1000 g for 10 minutes, collect the pellet, and wash it three times with 0.1 M sodium bicarbonate buffer (pH 8.3), finally resuspending the particles in the same buffer for storage. The polystyrene particles are labeled with fluorescent dyes using a chemical labeling method for subsequent experimental applications.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
