Abstract

A total of 12 previously undescribed eremophilane-type Sesquiterpenes agalleremonols A-L (1-12) were isolated from the agarwood of Aquilaria agallocha Roxb., and their structures were established based on extensive spectroscopic analyses. The absolute configurations of 1, 4, 5, and 12 were confirmed using single-crystal X-ray analysis, and the Others were determined through electronic circular dichroism (ECD) calculations. The protective activity of the isolated compounds against corticosterone (CORT)-induced damage in PC12 cells was assayed, and 2, 4, and 11 exhibited definite neuroprotective effects with survival rates of 82.14 %, 72.63 %, and 74.36 %, respectively. Flow cytometry and western blotting analyses indicated that the neuroprotective effect of 2 might be attributed to the alleviation of CORT-induced mitochondrial-dependent Apoptosis.
