Ionizable lipid-1 (compound II-10) is an ionizable lipid (pKa=6.16) that can be used to prepare lipid nanoparticles (LNP) with bilayer structure[1].

In Vitro

Ionizable lipid-1 hydrochloride can be protonated and positively charged at acidic pH. Ionizable lipid-1 hydrochloride electrostatically binds to negatively charged RNA molecules, thereby encapsulating RNA into nanoparticles. Ionizable lipid-1 hydrochloride will be protonated again once entering cells, interacts with the negatively charged endosome membrane. The interaction leads to the disruption of the endosome membrane, thereby releasing the RNA payload into the cytoplasm[2].
