Description

Hyaluronidase, Ovine testes is an endoglycosidase. Hyaluronidase, Ovine testes specifically degrades Hyaluronic acid (HY-B0633A) and Chondroitin sulfate (HY-B2162) by hydrolyzing β-glycosidic bonds in acidic mucopolysaccharides. Hyaluronidase, Ovine testes disperses follicular cells during fertilization by breaking down the hyaluronic acid-rich cumulus. Hyaluronidase, Ovine testes can be used in the study of fertility-related diseases[1][2][3].

In Vitro

Hyaluronidase inhibition test
Mix 10 μL of 8 mg/mL Hyaluronidase with the sample in a buffer solution (0.1 M acetate buffer, pH 3.6) and add it to a test tube. Incubate at 37 °C for 20 minutes.
(2) Add 20 μL of 12.5 mM calcium chloride to the mixture and incubate for another 20 minutes at 37°C.
(3) The activated Ca2+ mixture was treated with 50 μL of 2.4 μg/mL hyaluronic acid in the buffer and further incubated at 37°C for 40 minutes.
(4) Add 2 μL of 0.4 N sodium hydroxide and 20 μL of 0.4 N potassium tetraborate tetrahydrate, and then incubate in a water bath at 100°C for 3 minutes.
After cooling to room temperature, add 600 μL of DMAB solution to the mixture and incubate at 37°C for 20 minutes.
(6) Measure the absorbance at 585 nm using an microplate reader.
