Description

Taq DNA polymerase 2.0 is a thermostable DNA polymerase that can be used in PCR[1].

In Vitro

PCR system
2.5 μL 10x PCR Buffer; Up to 5 μL of whole blood template DNA; 2 μL 2.5 mM dNTP; 1 μL upstream primer (10 μM); 1 μL downstream primer (10 μM); 0.5 μL Taq DNA polymerase 2.0 The total volume of Nuclease-free water replenishment is 25 μL;
Note: The following reaction system takes 10×PCR Buffer as an example. If 10× high GC buffer is used, it is recommended to increase the dNTA concentration to 4 μL (in a 25 μL system).
Buffer component information:
10×PCR Buffer: 150 mM Tris, 50 0mM KCl,150 mM (NH4)2SO4, 20mM MgSO4, NP40 0.8% (PH 8.8);
10× High GC buffer: 200 mM Tris, 200 mM (NH4)2SO4, 15 mM MgSO4, NP40 0.8%, Betaine (HY-B0710) 1.5 M (PH 8.8).
PCR cycle
1) 95°C for 3 minutes Cycle once;
2) 94°C for 30 seconds; 55°C for 30 seconds; 72°C 45-90 seconds; Cycle 35 to 90 times
3) 72°C for 7 minutes; Cycle once.
