Reference procedures for the determination of cationic surfactants by photometric titration[1]:
1. Take an appropriate amount of cationic surfactant solution into a beaker, add deionized water, 0.5 mL of 1 mM Crystal violet, IND, and 4 mL of pH 2.2 buffer solution, then dilute to 40 mL.
2. Use a 0.1 mM potassium poly(vinyl sulfate) (PVSK) solution as the titrant and add it to the sample solution in successive portions.
3. After each addition, measure the absorbance of the solution at 590 nm using a spectrophotometer.
4. Before titration, the Crystal violet, IND does not react with the cationic surfactant, and the absorbance corresponds to the indicator itself. Before the equivalence point, the absorbance decreases slightly due to dilution by the titrant and ion association.
5. At the end point, the association of the Crystal violet, IND with the titrant causes a sharp decrease in its concentration, leading to a sudden drop in absorbance. The break point of the titration curve is taken as the end point.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
