Description

CellTracker CM-DiI (CM-DiI) is a lipophilic fluorochrome that can be used for tracking the migration of lymphocytes. CellTracker CM-DiI is feasible to detect labeled cells in histological sections (excitation 553 nm; emission 570 nm)[1][2][3].

In Vitro

Guidelines (The following is our recommended protocol, which is provided as a guide only and should be modified to suit your specific needs).
Using CM-DiI to label lymphocytes to track their migration[1]
1 Preparation of stock solution:
1) Dissolve 500 μg of CM-DiI in 250 μL of DMSO.
2 Preparation of working solution:
1) Transfer 7.5 mL of Hank's balanced salt solution preheated to 37°C to the DMSO solution containing CM-DiI and mix for a few seconds until the dye is evenly distributed.
3 Specific staining steps
1) Collect overnight lymph fluid and determine the cell concentration using a Coulter model ZM cell counter. Take a volume containing 1×109 lymphocytes, centrifuge at 4°C, 300×g for 10 min, discard the supernatant, and wash the pellet with HBSS.
2) Resuspend the washed cells in 2.5 mL HBSS and warm to 37°C.
3) Immediately transfer the dye working solution prepared above to the cell suspension and quickly mix by pipetting to a final cell concentration of 1×108 lymphocytes/mL.
4) Incubate the cells at 37°C for 2 min and then on ice for another 2 min.
5) Transfer the cells to a 50 mL centrifuge tube containing 40 mL 4°C HBSS, centrifuge and wash as above. Resuspend the pellet in 20 mL Lactated Ringer's solution, place the cell suspension on ice, and inject within 15-30 min.
