  3. AZ'320

AZ'320 is an ATP-competitive necroptosis (EC50 of 4.9 μM) inhibitor and a RIPK1 (pKD of 5.45) inhibitor. AZ'320 inhibits necroptosis by inhibiting RIPK1 phosphorylation. AZ’320 prevents mortality of the mice and rescues temperature and body weight in SIRS mice models. AZ'320 can be used for researchs of cancer and inflammation.

Description

AZ'320 is an ATP-competitive necroptosis (EC50 of 4.9 μM) inhibitor and a RIPK1 (pKD of 5.45) inhibitor. AZ'320 inhibits necroptosis by inhibiting RIPK1 phosphorylation. AZ’320 prevents mortality of the mice and rescues temperature and body weight in SIRS mice models. AZ'320 can be used for researchs of cancer and inflammation[1].

In Vitro

AZ’320 (0.1-10 μM, 4 h) dose-dependently inhibits the levels of RIPK1 Ser166 phosphorylation in I2.1 cells[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Western Blot Analysis[1]

Cell Line: I2.1 FADD−/− human Jurkat cells
Concentration: 01, 1, 5 and 10 μM
Incubation Time: 4 h
Result: Dose-dependently inhibited the levels of RIPK1 Ser166 phosphorylation.
In Vivo

AZ’320 (5 mg/kg, i.v., 15 min before TNF stimulation) prevents mortality of the mice and rescues temperature and body weight to a similar degree as Nec-1 (HY-15760) in SIRS mice models[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model: Systemic inflammatory response syndrome (SIRS) mice models (C57BL/6, male, 6-8 weeks)[1]
Dosage: 5 mg/kg
Administration: Intravenously injection, 15 min before 96 h of 0.5 mg/kg TNF stimulation
Result: Prevented mortality of the mice and rescued temperature and body weight to a similar degree as Nec-1 (HY-15760).

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
