Description

ATTO 488 is a hydrophilic rhodamine-based fluorescent dye with strong absorption and high fluorescence quantum yield. ATTO 488 is suitable for flow cytometry (FACS), fluorescence in situ hybridization (FISH) and most other labeling uses (Ex/Em: 500/520 nm)[1][2].

In Vitro

1. Preparation of ATTO 488 staining solution:
Prepare a 10 mM stock solution in DMSO and store at −80°C.
2. Labeling procedure:
Add a 2-fold molar excess of ATTO 488 to the purified protein.
Note: Labeling and spectral measurements should be performed quickly under green safe light or in the dark.
3. Incubation procedure:
Incubate for 2 hours at room temperature (RT) or overnight at 4°C with shaking in the dark.
4. Purification:
1) Remove free ATTO 488 using a NAP-5 size exclusion column.
2) Remove traces of free ATTO 488 by ultrafiltration.
5. Characterize the labeled protein by UV/Vis spectroscopy.
